abstract Retroviral vector comprising a nucleic acid comprising: a) one or two retroviral long terminal repeats (LTRs), b) a packaging signal, c) a rev sensitive element, d) a eukaryotic promoter, operably linked to e) a sequence of heterologous nucleotides, optionally in which the heterologous sequence comprises one or more marker genes, optionally selected from a 3-galactosidase gene, a hygromycin gene, a blastocidine gene, an MGMT gene, a neomycin gene, a gene of puromycin, a cytosine deaminase gene, a secreted alkaline phosphatase gene, a fluorescent protein gene, and combinations thereof; therapeutic genes; antiviral genes; antitumor genes; cytokine genes; genes encoding antigens; sequences that can be associated with host chromatin; sequences encoding a protein that can be associated with host DNA and host chromatin and vector nucleic acid; sequences encoding a protein that has DNA methylation activity, and combinations thereof, and optionally a post-transcriptional regulatory element, in which the retroviral vector lacks a 3'-polyurethane (3'-PPT) tract, optionally a functional cPPT.
