abstract | A method for identifying a peptide-cut enzyme inhibitor comprising: a. Perform a low kcat substrate assay by contacting each of a plurality of substrates with a target peptide cutting enzyme to identify at least one high kcat peptide substrate and at least one low kcat peptide substrate that is poorly cleavable by the target peptide cutting enzyme, in which the low kcat peptide substrate has a cleavage rate of less than 35% of the cleavage rate of the most highly cleaved substrate in the assay; and b. Perform a competitive binding assay to the active site of the enzyme using the target peptide cutting enzyme, at least one high kcat substrate identified in step a. and at least one low kcat substrate identified in stage a. and thus identify at least one low kcat substrate that is an inhibitor of active site binding, which inhibits the cleavage of at least one high kcat substrate by the target peptide cutting enzyme by binding to the active site of the enzyme target peptide cutting, thereby identifying a low kcat active site binding enzyme inhibitor comprising an amino acid nucleus or peptide nucleus. |
