abstract An immunogenic composition comprising a non-natural factor H (fHbp) binding protein derived from fHbp ID1 (having the sequence set forth in SEQ ID NO: 1) and having a sequence identity of at least 90% with SEQ ID NO: 1, in which the unnatural fHbp has an amino acid substitution at position R41, and in which said unnatural fHbp has a lower affinity for human H factor (fH) than fHbp ID1, and generates bactericidal antibodies against N. meningitidis; wherein the amino acid substitution in R41 is R41S or R41A; or wherein the composition comprises an adjuvant selected from aluminum hydroxide, aluminum phosphate, MF59 ™, an oil-in-water emulsion, monophosphoryl lipid A or a saponin; or wherein the composition comprises vesicles of the outer membrane of Neisseria meningitidis genetically modified to express fHbp; or wherein the composition comprises genetically modified Neisseria meningitidis microvesicles to express fHbp.
