abstract |
A covalent complex of von Willebrand factor (VWF) or variants thereof and variants of factor VIII, comprising a moiety that prolongs the half-life, provided that said factor VIII variants retain at least 10% of the biological activity of Non-mutated FVIII, in which factor VIII is modified so as to form a disulfide bridge with VWF, whereby factor VIII is modified by replacing a naturally occurring amino acid with a cysteine residue or inserting a Cysteine residue that forms a disulfide bridge with a cysteine residue in von Willebrand factor. |
