abstract A method for producing a multispecific antibody comprising a first polypeptide and a second polypeptide whose isoelectric points are different, wherein the method comprises the steps of: (A) modifying both or any one of a nucleic acid encoding amino acid residues of the first polypeptide and a nucleic acid encoding the amino acid residues of the second polypeptide, such that the difference between the isoelectric point of the first polypeptide and that of the second polypeptide will be 0.5 or more, in which the position (s) is modified (s) of said nucleic acid are selected from the group consisting of (i) and (ii) below: (i) at least one of positions 1, 3, 5, 8, 10, 12, 13, 15, 16, 19, 23, 25, 26, 39, 42, 43, 44, 46, 68, 71, 72, 73, 75, 76, 81, 82b, 83, 85, 86, 97, 105, 108, 110 and 112, Kabat numbering, in the variable region of the H chain, and (Ii) at least one of the positions 137, 196, 203, 214, 217, 233, 268, 274, 276, 297 , 355, 392, 419, and 435, EU numbering, in the constant region of the H chain; (b) culturing host cells to express nucleic acids; and (c) recovering the multispecific antibody from the host cell culture, using the difference at the isoelectric point, in which the first polypeptide and the second polypeptide comprise a variable region of the heavy chain.
