abstract |
A method of increasing the yield of a protein of interest in an animal or plant cell comprising transfection or transduction of said cell with at least one recombinant expression cassette encoding said protein of interest; and at least one other recombinant expression cassette encoding: (i) an XBP1 protein having the amino acid sequence of SEQ ID NO: 6 or a fragment thereof, wherein said protein or fragment thereof binds to a RPD element (ERPD) or to an ER stress response element (ERERE) of said cell; or (ii) an ATF6 protein having the amino acid sequence of SEQ ID NO: 9 or amino acid residues 1-366 of SEQ ID NO: 9, wherein said protein binds to an RPD element (ERPD) or to a ER stress response element (ERERE) of said cell; wherein the ratio of the total number of said at least one recombinant expression cassette to the total number of said at least one other recombinant expression cassette is at least 3: 1. |
