abstract | An in vitro method for determining whether a human subject has an altered DNA mismatch repair function, wherein said method comprises a) culturing primary fibroblasts derived from a sample of non-neoplastic constitutive tissue from said human subject, b) producing an extract of nuclear proteins from said primary fibroblast cells cultured from said human subject, c) providing extracts of nuclear proteins competent in MMR + / + MMR and lacking MMR - / - of cell lines as positive and negative controls, respectively, d) combining each of the nuclear protein extracts with at least one heteroduplex DNA substrate harboring mismatch in different vials, e) performing a mismatch repair assay on the nuclear protein extracts and f) quantitatively determining the capacity of the fibroblast cell nuclear protein extract primaries of repairing said substrate DNA molecule, in which a decrease in repair efficiency compared to the positive control is an indication of an impaired DNA mismatch repair function in the subject. |
