titleOfInvention | Particulate pharmaceutical composition |
abstract | Particulate pharmaceutical composition comprising: a drug that includes at least one biomachromolecule selected from the group consisting of a protein and a nucleic acid; and ** (See formula) ** a carrier composition comprising a block copolymer unit having a hydrophobic polymer chain segment and a water-soluble hydrophilic polymer chain segment, and a charged lipid carrying a charge opposite to the drug charge, in which the block copolymer unit is a compound represented by formula (I) or formula (II): in which ~ R1 and R3, independently of one another, are either an atom of hydrogen or a group represented by R8 (R9) CH (CH2) q-, in which R8 and R9: i) independently of one another, are a hydrogen atom, C1-6 alkoxy group, aryloxy group, aryl group C1-3-oxyl, cyano group, carboxyl group, amino group, C1-6 alkoxycarbonyl group, C2-7 acylamide group, tri- (C1-6 alkyl) -siloxy group, siloxy group, or silylamino group, ii) together between yes, they form an ethylenedioxy group or propylenedioxy group, which are either unsubstituted or substituted with C alkyl group 1-3, or iii) together with a CH group to which they are attached, form a formyl group, q is an integer from 0 to 10, R2 is a hydrogen atom, saturated or unsaturated C1-C29 aliphatic carbonyl group, or arylcarbonyl group, R4 is a hydroxyl group, saturated or unsaturated C1-C30 aliphatic oxyl group, or aryl- (lower alkyl) -oxyl group, R5 is -O- or -NH-, R6 is a hydrogen atom, phenyl group , benzyl group, group - (CH2) 4-phenyl, C4-C15 alkyl group that is either unsubstituted or substituted with amino group or carbonyl group, or sterol derived residue, R7 is a methylene group, n is a number integer from 55 to 4,600, x is an integer from 10 to 200, m is an integer from 0 to 200, where when m is one or more, the units (COCHNH) and the unit (s) (es) (COR7CHNH) are arranged in any order, and when m is two or more, R6 is selected for each amino acid unit independently of each other and arranged in the c block opolymer in a random order, provided that the hydrogen atoms do not represent 75% or more of R6, and is 1 or 2, L1 is a binding group selected from -NH-, -O-, -OZ-NH -, -CO-, -CH2- and -OZSZ-NH-, in which Z, independently of one another, means C1-C6 alkylene group, and L2 is a binding group selected from -OCO-Z-CO- and -NHCO-Z-CO-, in which Z is a C1-C6 alkylene group, and in which a plurality of the block copolymer units are arranged radially with the radially hydrophobic polymer chain segments radially inside and the segments radially hydrophilic polymer chain outside; the charged lipid being attracted to the hydrophobic polymer chain segment; and the drug is placed radially inside the hydrophobic polymer chain segments, whereby the drug is prevented from detaching from the particle. |
