abstract | A method for deriving a population of cells comprising cells expressing pluripotency markers, comprising the steps of: a. obtain a population of cells that express characteristic markers of the definitive endoderm lineage by differentiation of human embryonic stem cells and b. culturing the cells (a) under hypoxic conditions, on a tissue culture substrate that is not pretreated with a protein or extracellular matrix in a medium containing serum, activin A and a Wtn ligand, in which cells expressing markers of pluripotency express at least one of the pluripotency markers selected from the group consisting of ABCG2, crypto, FoxD3, connexin 43, connexin 45, Oct4, SOX-2, Nanog, hTERT, UTF-1, ZFP42, SSEA-3, SSEA- 4, Tra1-60 and Tra1-81. |
