abstract Method for examining a compound to determine if the compound is a high specificity proteasome desubiquitination inhibitor, the method comprising contacting the compound with human 19S regulatory particles (RP 19S) of 26S proteasome and determining whether the compound inhibits the activity of deubiquitination enzymes (DUB) UCHL5 and USP14, and an additional step of determining whether the compound affects the activity of associated non-proteasomal DUB enzymes, in which the inhibition of the activities of UCHL5 and USP14 and the unaffected activity of Associated non-proteasomal DUB enzymes indicates that the compound is a high specificity proteasome desubiquitination inhibitor.
