abstract | A pyrosequencing method of a polynucleotide molecule, said method comprising the steps of: providing a rotating platform having at least one open well to contain at least one support surface, said open well being configured or sized such that a reagent deposited therein is removable by centrifugation from said open well and out of said platform by sufficient rotation of said platform; providing at least one said support surface in the form of a magnetic particle in each said open well, wherein said support surface is adapted to immobilize a polynucleotide molecule or has a polynucleotide molecule immobilized thereon; if there is no polynucleotide molecule already immobilized thereon, immobilize a polynucleotide molecule on said magnetic particle; tuning an oligonucleotide primer into a single chain of said polynucleotide molecule; dispense in each open well cited from an external point of said platform, a series of pyrosequencing reagents, wherein after one or more dispensing stages mentioned, said platform is sufficiently rotated such that any residual reagent mentioned or not reacting is substantially extracted by centrifugation from each said open well and out of said platform, wherein during rotation, each said magnetic particle is magnetically held inside each said well; performing an assay with respect to the presence of a pyrophosphate group in each said well, said test step comprising detecting a light signal in each said open well; and repeating said dispensing and testing steps, thereby sequencing said polynucleotide molecule. |
