abstract A method of separating a target oligonucleotide from a mixture of the target oligonucleotide and one or more impurities comprising: providing a two-phase liquid-liquid chromatography system mobile phase-stationary phase comprising a first liquid mobile phase and a liquid stationary phase containing at least one exchange substance that binds with the possibility of separation to the target oligonucleotide; causing the first liquid mobile phase to transport the target oligonucleotide in a flow in relation to and in contact with the liquid stationary phase in the column of a liquid-liquid chromatography apparatus so that the target oligonucleotide is attached to the exchange substance in the phase liquid stationary; then, moving the target oligonucleotide from the liquid stationary phase to a second liquid mobile phase that flows in relation to and in contact with the stationary phase through the column by means of a displacement substance capable of displacing the target oligonucleotide from the phase stationary liquid to the second mobile phase, characterized in that the exchange substance is a salt of a secondary, tertiary or quaternary organic amine with a counter anion, of general formula: R1R2R3R4N + X in which through the secondary, tertiary and quaternary sequence, respectively, two, three or four of the groups R1, R2, R3 and R4 are independently C1-20 alkyl or substituted alkyl such as fluoro- or trifluoromethyl alkyl or benzyl and the others are hydrogen, and X- is a halide anion.
