abstract |
A stable affinity chromatography ligand in alkaline medium, comprising two or more C domains of Staphylococcus Protein A (SpA) bound to a solid support by a multipoint binding, in which at least one domain comprises: a) a sequence of amino acids set forth in SEQ ID NO: 11 or an amino acid sequence encoded by the amino acid sequence set forth in SEQ ID NO: 5; or b) an amino acid sequence that has a sequence identity of at least 95% with the amino acid sequence set forth in SEQ ID NO: 11 or an amino acid sequence that has a sequence identity of at least 95% with the sequence of amino acids encoded by the amino acid sequence set forth in SEQ ID NO: 5; and wherein each C domain comprises a substitution of glycine at position 29 with an amino acid other than alanine or tryptophan and in which additionally the ligand retains at least 95% of its binding capacity after 5 hours of incubation in NaOH 0.5 M. |
