abstract | A method of amplifying a nucleic acid target sequence template contained in a sample, comprising: contacting the sample with an amplification reaction mixture containing a primer complementary to the nucleic acid target sequence template and an agent nucleic acid destabilizer comprising at least one of DMSO, formamide and betaine, at a concentration of 8-15 percent by volume, the primer having a melting temperature> 55 ° C; oscillating a reaction temperature between a higher denaturation temperature and a lower hybridization temperature, in which a change in temperature is not greater than 20 ° C for a plurality of temperature cycles, the upper denaturation temperature being sufficiently high for completely denature the template; and amplify the template of the nucleic acid target sequence. |
