abstract | Procedure for the detection or quantitative determination of an inhibitor of a hemostatic system peptidase in a sample, the hemostatic system peptidase being a coagulation factor, selected from the group consisting of factor IIa (thrombin), VIIa, IXa, Xa, XIa as well as fragments and mutants of the mentioned peptidases, which show at least 80% of the capacity of the mentioned peptidases to enzymatically convert their coagulation system substrates, and the sample is selected from blood, plasma, serum, cerebrospinal fluid, urine, sweat or other fluid, comprising contacting the inhibitor in the sample with the peptidase coupled to a polymer of the hemostatic system, and the polymer is a polyalkylene glycol or a copolymer, comprising alkylene glycol units, and wherein the The hemostatic system peptidase, due to polymer coupling, loses its reaction capacity in the coagulation system, but, at at the same time it can still neutralize inhibitors of the coagulation system with a molecular mass of between 100 and 2,500 Da and can enzymatically convert substrates of the coagulation system with a molecular mass of between 100 and 2,500 Da, or (i) the measurement of activity of the polymer-coupled peptidase after contact with the inhibitor in the sample, and the comparison of the activity of the coupled peptidase after contacting the sample with one or more reference values, to establish a possible inhibition of activity; or (ii) the measurement of the coagulation time of the sample after contact with the coupled peptidase, and the comparison of the coagulation time with one or several reference values, to establish a possible variation of the coagulation time. |
