abstract | A sample analysis method, comprising: subjecting a reaction mixture comprising: PCR reagents to amplify a nucleic acid target, and flap cleavage reagents to carry out a flap cleavage assay on said nucleic acid target, to the following thermocycling conditions: a first series of 5-15 cycles of: i. a first temperature of at least 90 ºC; ii. a second temperature of 60ºC to 75ºC; iii. a third temperature of 65 ºC to 75 ºC; followed by: a second series of 20-50 cycles of: i. a fourth temperature of at least 90 ºC; ii. a fifth temperature that is at least 10 ° C lower than said second temperature; iii. a sixth temperature of 65 ºC to 75 ºC; no additional reagent being added to said reaction between said first and said second series of cycles and the segmentation of a flap probe being measured in each cycle of said second series of cycles, and said method comprising measuring the segmentation of said flap probe while the reaction mixture is at said fifth temperature. |
