A pharmaceutical composition formulated for local application to a surface of an epithelial tissue barrier, said composition comprising a soluble antigen-binding polypeptide and a pharmaceutically acceptable carrier, diluent, or excipient, wherein said antigen-binding polypeptide can cross a cornea mammal intact in less than 8 hours in the absence of a penetration enhancer, and wherein said antigen-binding polypeptide is a single-chain antibody (scFv) and comprises: (a) a variable domain framework region of light chain (VL) of at least 85% similarity to a VL lattice region selected from the group consisting of SEQs. ID NO: 1, 2, 3, 4, 5, 6 and 7; and a heavy chain variable domain (VH) framework region of at least 85% similarity to a VH framework region selected from the group comprising SEQ.
