Chimeric antigen receptors with an optimized hinge region |
abstract |
A protein comprising (i) a signal peptide; (ii) a specific target recognition domain; (iii) a linker region, which connects domain (ii) and domain (iv), in which the linker region does not contain a cysteine residue or moieties and is selected from any of the following: the amino acid sequence of SEQ ID NO. 2, an amino acid sequence with at least 95% sequence identity with SEQ ID NO. 2, with the proviso that amino acid residue 48 is not a cysteine and is a serine, and an amino acid sequence that differs by one, two or three amino acid residues from the amino acid sequence of SEQ ID NO. 2, with the proviso that the remainder of amino acid 48 is not a cysteine and is a serine; and (iv) an effector domain comprising a transmembrane region and one or more intracellular signaling domains, in which the effector domain (iv) comprises or is a fusion of the human CD28 transmembrane and intracellular domain with the intracellular chain domain zeta of human CD3. |
