abstract | An in vitro method for diagnosing an autoimmune disorder, where the open transglutaminase form or antigenetically active fragments of the open transglutaminase form are used for detection of the presence of open transglutaminase specific autoantibodies or antigenically active fragments of the open form transglutaminase INASE, in which antigenically active fragments of the open transglutaminase form are peptides after reaction with an inhibitor comprising a main chain, a peptide backbone or a peptidomimetic backbone and a reactive group of thiol capable of covalently bonding to the thiol group of cysteine at the active site of the respective transglutaminase and the antigenically active fragments have an amino acid residue arrangement that forms part of the surface of the open transglutaminase form exposed to activation of enzyme and wherein the antigenically active fragments are capable of eliciting an antibody response against epitopes, which are only accessible to antibodies after binding of an inhibitor. |
