abstract |
A method of conjugating a polysalic acid (PSA), which contains an active aminooxy group, to an oxidized carbohydrate moiety of a blood coagulation protein, which comprises contacting the oxidized carbohydrate moiety with an activated PSA under conditions that allow conjugation; wherein said blood coagulation protein has biological activity of Factor IX (FIX) or biological activity of Factor VIII (FVIII); wherein said carbohydrate moiety is oxidized through incubation with a buffer comprising an oxidizing agent selected from the group consisting of sodium periodate (NalO4), lead tetraacetate (Pb (OAc) 4) and potassium perrutenate (KRuO4); and that an oxime bond is formed between the oxidized carbohydrate moiety and the active aminooxy group in the PSA. |
