A method of amplifying a target nucleic acid, comprising: a. denature the target nucleic acid; b. hybridize oligonucleotide primers with the target nucleic acid; and c. extend the hybridized oligonucleotide primers; wherein the stages of denaturation, hybridization and spreading are carried out by thermal cycles within 10 ° C of the melting temperature of the target nucleic acid, and within the overlap between the melting temperatures of the oligonucleotide primers and the temperature of fusion of the target nucleic acid; and wherein the oligonucleotide primers are designed to have a melting temperature that is within about 10 ° C of the melting temperature of the target nucleic acid. |
