abstract Method of in vitro detection of an enzyme of a third generation cephalosporin resistant microorganism from a biological sample, said enzyme being selected for detection in the group consisting of extended spectrum b-lactamases, cephalosporinases and carbapenemases, said method comprising stages consisting of: a1) concentrating the microorganisms present in the biological sample, optionally after a step a0) of cultivation of the microorganisms; b1) suspending the microorganisms concentrated in step a1) in a solution comprising at least one chromogenic substrate capable of releasing a chromophore after hydrolysis by the enzyme for detection, said chromogenic substrate being the compound HMRZ-86 (acid trifluoroacetate (7R) -7- [2- (aminothiazol-4-yl) - (z) -2- (1-carboxy-1-methylotoxyimino) acetamido] -3- (2,4-dinitroestiryl) -3-cefem-4 -carboxylic, isomer E) or a salt thereof; c1) detect the possible release of the chromophore obtained in step b1); the chromophore release being detected in step c1) indicative of the presence of the enzyme for detection and further comprising said procedure, when the biological sample is a sample containing blood or red blood cells: - a stage of lysis of the red blood cells present in the biological sample before stage b1) of suspension or before stage a1) of concentration, and / or - a stage a ') of preparation of the biological sample before stage a1) of concentration of microorganisms, comprising the step a '): (i) the agglutination of the red blood cells, and (ii) the separation of the red blood cells from the microorganisms present in the sample.
