Beta- (1.42) amyloid oligomers, derivatives thereof and antibodies thereto, methods for preparing them and using them |
abstract |
Oligomer of the β-1-42 truncated N-terminally soluble amyloid in physiological saline as opposed to rather globular fibrillar forms comprising fragments of Aβ (X-42), in which X represents a number from 10 to 24, the oligomer can be achieved according to a method comprising: (a) the detergent can act on a monomeric amyloid β (1-42) protein to form an Aβ oligomer (1-42) soluble in rather globular physiological saline solution unlike fibrillar forms; (b) the effect of the detergent is reduced and oligomer A of Aβ (1-42) is re-incubated to form an oligomer B of Aβ (1-42) soluble in physiological saline rather globular unlike forms fibrillar; and (c) oligomer B of Aβ (1-42) is treated with protease, the detergent being a compound of the formula RX, wherein the moiety R optionally represents branched alkyl with 6 to 20 carbon atoms or optionally alkenyl branched with 6 to 20 carbon atoms, and the remainder X represents an acidic group or its salt, and the oligomer B of Aβ (1-42) having an apparent molecular weight in the SDS gel electrophoresis of approximately 38 kDa and / or approximately 48 kDa. |
