abstract |
A type III fibronectin polypeptide monobody (Fn3) comprising a plurality of sequences of the ß-strand domain of Fn3 that are linked to a plurality of sequences in the loop region, in which one or more sequences in the loop region of the monobody vary by deletion, insertion or substitution of at least two amino acids from the corresponding sequences of the natural Fn3 loop region; wherein the Fn3 polypeptide monobody comprises a stabilizing mutation of at least one amino acid residue compared to a natural Fn3 molecule, in which the stabilizing mutation is a substitution of at least one amino acid residue that is involved in an electrostatic interaction unfavorable, in which the stabilizing mutation is a substitution of at least one Asp 7, Asp23 or Glu 9 with another amino acid residue, wherein said stabilizing mutation increases the melting point of the Fn3 polypeptide monobody by more than 0.1 ° C compared to an Fn3 polypeptide monobody that is identical except for the change. |
