abstract | A method of detecting antibody-drug conjugated compounds comprising: (i) processing by immunoaffinity membrane selection a biological sample that has been collected from a biological source in contact with a antibody-drug conjugate compound having Formula I previously : Ab- (LD) p I in which Ab is an antibody that binds to one or more antigens associated with tumors or cell surface receptors; D is a drug residue selected from a maitansinoid or auristatin; L is a linker covalently linked to Ab and covalently linked to D, wherein L is formed from a linker reagent selected from N-succinimidyl 4- (2-pyridylthio) propanoate (SPDP), succinimidyl-4- (N- maleimidomethyl) cyclohexane-1-carboxylate (SMCC) and N-succinimidyl-4- (2-pyridylthio) pentanoate (SPP), or L is selected from maleimidocaproyl (MC), maleimidopropanoyl (MP) and maleimidocaproyl-valine-citrulline-paraaminobenzyloxycarbonyl (benzyloxycarbonyl) MC-vc-PAB); and p is 1, 2, 3, 4, 5, 6, 7 or 8; wherein a test sample is formed by contacting the biological sample with an immunoaffinity membrane comprising an immobilized antigen and elution of the test sample; (ii) applying the test sample to a separation medium to effect separation of more than one constituent sample wherein a constituent of the separated sample comprises an antibody fragment of an antibody-drug conjugate compound having Formula I, or an antibody fragment or metabolite thereof, and in which p is 0, 1, 2, 3, 4, 5, 6, 7 or 8; and (iii) establish the mass or ratio between the mass and the charge and quantify one or more separate sample constituents that is an antibody fragment or an antibody-drug conjugate compound having Formula I or a metabolite thereof , where p is 0, 1, 2, 3, 4, 5, 6, 7 or 8, by mass spectrometry with single ion monitoring (SIM). |
