abstract | Method comprising: (a) incubating isolated cells from a breast tumor with an in vitro anticancer drug, (b) lysing the incubated cells to produce a cell extract, (c) detecting an activation state of one or more analytes in the cell extract using an assay comprising a plurality of dilution series of specific capture antibodies of one or more analytes, in which the capture antibodies are immobilized on a solid support, and in which the assay comprises: (i ) incubating the cell extract with a plurality of dilution series of capture antibodies to form a plurality of captured analytes, (ii) incubating the plurality of analytes captured with detection antibodies comprising a plurality of antibodies independent of the activation state and a plurality of activation-dependent antibodies specific for the corresponding analytes in order to form a plurality of analytes detectable captured os, in which the independent activation state antibodies are labeled with glucosidase oxidase, in which the glucose oxidase and the independent activation state antibodies are conjugated with a sulfhydryl activated dextran molecule, in which the antibodies dependent on the activation state are marked with a first member of a signal amplification partner and in which glucose oxidase generates an oxidizing agent that is channeled and reacts with the first member of the signal amplification partner, (iii) incubate the plurality of captured analytes detectable with a second member of the signal amplification pair to generate an amplified signal, and (iv) detecting the amplified signal generated from the first and second members of the signal amplification pair, and (d ) determine whether the anticancer drug is suitable or not for the treatment of breast tumor by comparing the Activation status of one or more analytes with a reference activation profile generated in the absence of the anticancer drug, in which the method is intended to select an anticancer drug suitable for the treatment of a breast tumor, in which they are isolated cells from the mammary tumor of a subject who has not received treatment with the anticancer drug, and in which the reference activation profile provides the activation status of one or more analytes in breast tumor cells before treatment with the anticancer drug. |
