A polynucleotide encoding a neurotoxin polypeptide comprising a light chain, a linker, and a heavy chain, wherein the linker is a modified linker comprising a heterologous amino acid sequence that confers at least one physicochemical property to the polypeptide that allows separation of unprocessed or partially processed neurotoxin polypeptides of processed neurotoxin polypeptides, said heterologous amino acid sequence flanked at the N- and C-terminus by a protease recognition and cleavage site, wherein the neurotoxin is a botulinum neurotoxin. |
