abstract | A method of producing a transformed plant that does not contain a selectable marker in its genome, which comprises (a) infecting plant cells with Agrobacterium containing a single binary vector comprising (i) a transfer DNA containing a gene of interest and (ii) an expression cassette containing a plant biosynthetic hormone gene positioned outside the transfer DNA to express a cytokinin biosynthetic gene; (b) culturing the cells in a hormone-free medium to produce shoots; (c) identify an outbreak that has genomic DNA that contains the transfer DNA but not the cytokinin biosynthetic gene expression cassette by selection without the use of antibiotic resistance genes; and (d) cultivate a plant from the identified outbreak of (c), where the plant is a transformed plant that does not contain a biosynthetic gene of the plant hormone in its genome. |
