abstract |
A composition comprising a substantially homogeneous mixture of duplex-forming oligonucleotides, wherein the mixture of duplex-forming oligonucleotides is formulated in a low salinity buffer and includes a solute, wherein at least 50% of the duplex-forming oligonucleotides they are present in monomeric or dimeric form and wherein the duplex-forming oligonucleotides are immunostimulatory oligonucleotides comprising: a TLR activation domain in 5 'and at least two palindromic regions, a palindromic region being a palindromic region in 5' of at least 6 nucleotides in length and connected to a 3 'palindromic region of at least 8 nucleotides in length both directly and through a spacer, which is a nucleic acid that is 1-50 nucleotides in length or a non-nucleotide spacer, where the oligonucleotide includes at least one YpR dinucleotide, and wherein the oligonucleotide is not T * C_G * T * C_G * A * C_G * T * T * C_G * G * C * G * C_G * C * G * C * C * G (SEQ ID NO: 335). |
