abstract | A method of identifying the origin of a metastasis of unknown origin that comprises the steps of a. Measure the Biomarkers associated with at least six different carcinomas in a sample containing metastatic cells where the Biomarkers are expression levels of at least the following Marker genes: SP-B, TTF1, DSG3, PSCA, F5, HPT1, PSA, MGB / MG, WT1 and PDEF; b. combine the data of the Biomarkers in an algorithm where the algorithm i. Normalize the Biomarkers against the reference; and ii. imposes a cut-off point that optimizes the sensitivity and specificity of each Biomarker, weighs the prevalence of carcinomas and selects a tissue of origin; C. determine the origin based on the highest probability determined by the algorithm or determine that the carcinoma is not derived from a particular set of carcinomas; and d. optionally measure specific Biomarkers for one or more additional different carcinomas, and repeat steps c) and d) for additional Biomarkers. |
