A method of providing DNA fragments derived from an archived sample, comprising: obtaining an archived sample comprising DNA and paraffin and optionally a binding agent; (a) subjecting the archived sample directly to a lysis stage with a protease, wherein the paraffin is liquefied by heating to provide an amount of DNA treated with accessible protease; (b) inactivate the protease by heat, addition of a protease inhibitor, or both; and (c) treating the DNA with a bisulfite reagent without a previous step of DNA extraction. |
