abstract | A method for measuring the fraction of viable cells in a cell population maintained in a cell culture medium, the method comprising: (a) detecting a stable enzymatic activity to cell death (Y) in a portion of a conditioned cell culture medium which does not contain cells of the cell population; (b) destroy the integrity of the cell membrane in a portion of the cell culture medium containing population cells; (c) detecting a stable enzymatic activity to cell death (X) in the cells and conditioned medium of said portion of the cell culture medium containing cells of the cell population; and (d) compare the level of stable enzyme activity to cell death (X) in the cells and conditioned medium of the portion of the cell culture medium containing cells with the level of stable enzyme activity to cell death (Y) in the portion of the conditioned medium of the cell culture that does not contain cells; in which the fraction of viable cells in the cell population can be calculated according to the formula: in which c 1. and f is the fraction of the total conditioned medium present in the portion of the conditioned medium of the cell culture that does not contain cells of the cell population. |
