abstract | The invention comprises compounds of the formula <FORM:0912131/IV (b)/1> (wherein R1 is a phenyl, 2-thienyl, cyclohexyl, halophenyl, lower alkyl-phenyl or lower alkoxy-phenyl, R2 is phenyl, cyclohexyl, 2-thienyl or cyclopentyl, R4, R5, R6 and R7 are hydrogen or alkyl, X and Z are straight or branched lower-alkylene groups having at least 2 carbon atoms in a straight chain between the nitrogen and the ester group, and A is hydroxy, halogen or an acyloxy group derived from benzoic acid, a phenylalkanoic acid of at most 8 carbon atoms or from a lower aliphatic carboxylic acid, the word "lower" referring to groups containing up to 6 carbon atoms) and acid-addition salts thereof and their preparation by reacting the appropriate di-hydroxyalkyl-piperazine either with a compound of the formula <FORM:0912131/IV (b)/2> (wherein A is a halogen or an acyloxy group and B is a reactive halogen) and, when required, hydrolysing A to form the dihydroxy-derivative, or with a compound of the formula <FORM:0912131/IV (b)/3> (wherein R3 is a hydrocarbon group). Examples are given and inorganic and organic salt-forming acids are referred to. 1 : 4-Bis-(hydroxyalkyl)-piperazines are prepared by reacting a piperazine or a 1-hydroxyalkyl-piperazine with a lower alkylene oxide. Examples are given. The substituted piperazines of the invention may be formulated as pharmaceutical compositions such as tablets, capsules and solutions. They are stated to antagonize the psychotogenic effects produced by N-alkyl-3-piperidyl benzilates and to produce relaxation without hypnosis, and relief from anxiety and restlessness. U.S.A.
