abstract A stable affinity chromatography ligand in alkaline medium, comprising two or more C domains of Staphylococcus Protein A (SpA) bound to a solid support by a multipoint binding, wherein each C domain comprises a glycine substitution in the position 29 with an amino acid other than alanine, threonine or tryptophan, in which domain C comprises an amino acid sequence with an identity of at least 90% with the amino acid sequence of SEQ ID NO: 1 or is encoded by the sequence of nucleic acids of SEQ ID NO: 5, except for the replacement of glycine at position 29, and wherein additionally the ligand retains at least 95% of its binding capacity after 5 hours of incubation in 0.5 M NaOH .
