Method for preparing a recombinant virus comprising a gene of interest, wherein said method comprises a) providing a packaging cell line that is transfected with a retroviral vector comprising a polynucleotide comprising a gene of interest, and a vector comprising a gene that encodes a targeting molecule; wherein said targeting molecule comprises an alphavirus glycoprotein that selectively and specifically recognizes the DC-SIGN (non-integrin binding protein of ICAM-3 [type 3 intracellular adhesion molecules] specific for dendritic cells); wherein said glycoprotein is an alpha2 virus E2 glycoprotein that is mutated at a heparan sulfate binding site to decrease or eliminate its heparan sulfate binding capacity, b) culturing the packaging cell line; and c) recovering the recombinant virus from the packaging cell line. |
