abstract | A storage-stable, substantially anhydrous topical composition comprising a homogeneous mixture of (a) a therapeutically effective amount of calcipotriol in dissolved form; (b) a solvent mixture consisting practically of a fatty acid ester solvent selected from the group consisting of glyceryl esters, for example fatty acid triglycerides, C10-18 alkanoic or alkenoic acid isopropyl esters, for example isopropyl myristate, isopropyl palmitate, isopropyl isostearate, isopropyl linoleate, isopropyl monooleate, or propylene glycol esters, for example, propylene glycol dipelargonate, or mixtures thereof; a fatty alkyl ether co-solvent which is a compound of the general formula IH (OCH2C (R2) H) xR1, wherein R1 is a linear or branched C1-20 alkyl chain, each R2 being individually hydrogen or CH3, and x is an integer of 2-60; and a lipophilic penetration enhancer selected from the group consisting of N-alkylpiperidone, Nalkylpyrrolidone, such as N-methylpyrrolidone or N-hydroxyalkylpyrrolidone, or dimethylacetamide, dimethylsulfoxide or 2-pyrrolidone; wherein the ratio of fatty acid ester solvent to fatty alkyl ether co-solvent to penetration enhancer is in the range of 50:25:25 to 75:10:15; said solvent mixture is included in the composition in an amount that is sufficient to effectively dissolve said calcipotriol, and (c) a substantially anhydrous pharmaceutically acceptable carrier. |
