Mix the components of stages ad and provide conditions that allow the primer to hybridize with the template - in which the first oligonucleotide is an oligonucleotide between 5 and 60 nt in length, comprising a TINA monomer of the formula Z, in which Z is described by the general structure: X-L-I1-C-I2 -in which X is a structural monomer unit that can be incorporated into the structure of an oligonucleotide or a nucleotide analogue, or PNA, or analogues of PNA, L it is a linker, I1 is a first intercalator comprising at least an essentially flat conjugate system, which is capable of co-stacking with nucleotide bases of DNA, RNA or analogs thereof, C is an optional conjugate and I2 is a second intercalator that it comprises an essentially flat conjugate system, which is capable of co-stacking with nucleotide bases of DNA, RNA or analogs thereof f. Under conditions that allow primer extension, prolong the first oligonucleotide hybridized with the template.
