abstract A deep density gradient filter device (100) for primary or secondary clarification of processed biopharmaceutical fluids, comprising in order: a pre-filtration zone (20) comprising layers (21,22) of filtration material, said zone of prefiltration (20) is capable of retaining particles within a first size distribution range; a primary filtration zone (30) comprising at least one fibrillated fiber layer containing material, said primary filtration zone is capable of retaining particles within a second size distribution range, said second size distribution range is smaller that said first size distribution interval; and a polishing zone (40) comprising layers (41 to 45) of filter material, said polishing zone (40) is capable of retaining particles within a third size distribution range, said third size distribution range is smaller that said second size distribution range, wherein the device is capable of filtering a fluid containing a particle distribution in the range of 25 microns to 0.2 microns; wherein said first size distribution range is 25 microns to 1 micron, said second size distribution range is 1 micron to 0.3 microns, and said third size distribution range is 0.3 microns to 0.2. microns; and where the retention capacities of the pre-filtration zone (20), the primary filtration zone (30), and the polishing zone (40) are set at a fluid velocity of at least 100 cm / hr and a hydraulic permeability greater than 1.45 cm / h / kPa (10 cm / hr / psi), where the layers of filter material in said filter device have a total summed depth of at least 1.27 cm (0.5 in. ), and where the ratio of the frontal area to the volume of the deep density gradient filter device (100) is 20 to 80 m-1.
