abstract Method for the production of a gene product of interest, the method comprising the steps of: a) providing an insect cell comprising: i) a first expression cassette comprising a nucleotide sequence encoding the gene product of interest that it is operatively linked to a promoter capable of activating gene product expression in the insect cell, and in which the first expression cassette is flanked on either side by inverted terminal repeat nucleotide (ITR) sequences of adeno-associated virus (AAV); ii) at least a second expression cassette comprising a nucleotide sequence encoding at least one AAV Rep protein that is operably linked to a promoter capable of activating expression of Rep proteins in the insect cell; b) culturing the insect cell defined in a) under a condition that leads to the expression of the first and second expression cassettes, and c) optionally recovering the gene product of interest, in which the insect cell does not comprise a Parvoviral Cap protein or a nucleotide sequence that encodes a Parvoviral Cap protein, and in which the first and second expression cassettes are comprised in the same one or two separate baculoviral vectors.
