abstract | A method for the conversion of acetolactate to dihydroxyisovalerate, comprising: a) providing a microbial host cell comprising a genetic construct encoding a polypeptide having a specific activity of cetol-acid reductoisomerase greater than the specific activity of a ketol-acid reductoisomerase E coli, in which said polypeptide has a specific activity of cetol-acid-reductoisomerase greater than 1.1 μmol / min / mg based on the purified protein, measured by the NADPH consumption test, executed under the following conditions: i) pH of about 7.5; ii) a temperature of approximately 22.5 ° C; and iii) potassium greater than about 10 mM; and b) contacting the host cell of a) with acetolactate, in which 2,3-dihydroxyisovalerate is produced. |
