abstract | A continuous method for preparing active agent-containing polymeric microspheres, comprising: a) forming a dispersed phase containing active agent and polymer; b) provide a continuous phase; c) continuously introducing said dispersed phase into a reactor vessel at a dispersed phase feed rate of from 4 ml/minute to 400 ml/minute, and said continuous phase into said reactor vessel at a continuous phase feed rate of 1,000 ml/minute to 20,000 ml/minute, wherein said continuous phase and said dispersed phase are introduced into said reactor vessel at proportions in a ratio of 40:1 to 200:1, the vessel including means for forming an emulsion, and forming a emulsion of the disperse phase in the continuous phase; d) introducing said dispersed phase into said continuous phase in a mixing zone where a shear force or turbulence is caused by a non-static mixer in said reactor vessel, and rotating an impeller of said non-static mixer at a higher mixing intensity than 5,000 revolutions per minute, such that said polymer of the dispersed phase solidifies in said continuous phase in less than 5 seconds after being introduced into said mixing zone, forming a suspension of microspheres; and e) continuously transporting said microsphere suspension from said reactor vessel to a solvent removal vessel to remove solvent from said continuous phase. |
