abstract | Procedure for the preparation of a conjugate, which includes the following components: one or more polypeptide molecules (I) based on human ubiquitin, and, covalently linked with it, one or more functional components (II) that are chosen from the set formed by polypeptides and proteins, organic polymers, sugars, low molecular weight substances, peptides, as well as derivatives of these substances, realizing that, after the coupling of (I) to (II), the functionality of all the components remains preserved, and realizing that the procedure comprises the following steps: a) Provision of human ubiquitin; b) Provision of a ligand, which is chosen from the set consisting of some (poly) peptides / proteins; c) Selection of amino acids 2, 4, 6, 62, 63, 64, 65 and 66 of human ubiquitin; d) Modification of the selected amino acids by substitution mediating conservation of the ubiquitin-like folding pattern; e) Contacting the modified protein with the ligand made available in step b); f) Determination of those proteins, which are suitable for a specific binding to ligands with a KD dissociation constant> = 10-5 M or less, detecting binding activity using the ELISA technique or a MRI superficial plasmons; g) Coupling of components (I) and (I) in a region located outside the superficially exposed region of the ß-folded sheet of the polypeptide molecule (I), which is intended for specific binding to the ligand, through terminal fusion of peptides with (I), or through cysteine or lysine side chains in (I); h) Isolation of the conjugate; and i) Detection of the functionality of both components of the conjugate. |
