abstract | Method for measuring the concentration of factor VIIa-antithrombin complexes in a plasma sample of a patient comprising the following steps: mixing an amount of said plasma sample with a primary capture antibody fixed to a solid phase, said primary antibody being characterized because it has a binding capacity a) to the factor VIIa part or b) to the antithrombin part of the factor VIIa-antithrombin complexes that may be present in said plasma sample, and incubated the mixture under conditions that promote the union between said capture antibody and said factor VIIa-antithrombin complexes that could be present in said plasma sample to form bound VIIa-antithrombin complexes and an unbound mixture; extract said unbound sample; mixing said bound factor VIIa-antithrombin complexes with a secondary antibody characterized in that it presents a binding capacity a) to the antithrombin part b) to the factor VIIa part, respectively, of said bound factor VIIa-antithrombin complexes to form complexes of bound secondary VIIa-antithrombin antibody and a mixture of unbound secondary antibody; extract any mixture of unbound secondary antibody; determine the amount of bound secondary antibody; and comparing the amount of bound secondary antibody with a standard amount of bound bound secondary antibody for bound factor VIIa-antithrombin complexes of a known concentration. |
