abstract | A method for extending and selectively immobilizing a polynucleotide, which comprises the operations of: a) immobilizing a first portion of said polynucleotide on a surface; b) applying to said polynucleotide a force capable of extending said polynucleotide; and c) putting a second portion of said polynucleotide in contact with a molecule, wherein said molecule comprises a first component capable of binding to the second portion of said polynucleotide and a second component capable of selectively binding to said surface, under conditions under which which said molecule selectively binds to the second portion of said polynucleotide and selectively binds to said surface, thereby immobilizing said polynucleotide in an extended state. |
