abstract A method for modifying the methylation pattern of a target cell DNA sequence, comprising the following steps: a) providing a suitable polynucleotide for induction of RNA-directed DNA methylation of said plant cell, said polynucleotide comprising a sequence which is complementary to the target DNA sequence of said plant cell and that is inserted into a plant intron, b) introduction of said polynucleotide into said plant cell, c) expression of said polynucleotide in said transformed cell, thereby inducing methylation of directed DNA by RNA, and d) determination of the methylation pattern of said DNA target sequence in said plant transformed cells.
