abstract | Method for testing homocysteine (Hcy) in a sample without chromatographic separation, the method comprising: a) contacting a sample that contains or is suspected of containing Hcy, with a Hcyun Hyss converter enzyme cosustrate in a reaction Hcy conversion to form an Hcy conversion product and an Hcy cosustrate conversion product, in which the Hcy cosustrate is S-adenosylmethionine (SAM), the Hcy converter enzyme is the S-methyltransferase-dependent homocysteine S-adenosylmethionine (SAM), the Hcy conversion product is methionine (Met) and the Hcyes cosustrate conversion product S-adenosyl-L-homocysteine (SAH), b) contacting the mixture with an adenosylhomocysteine nucleosidase to convert SAH into adenine and Sribosil-homocysteine, c) contacting the mixture with an S-ribosyl-homocysteinase to cycle the S-ribosyl-homocysteine enhomocysteine, and d) assess the amount of adenine in the sample, in which the amount of Hcy in the sample is determined from the amount of adenine. |
