abstract | Method of preparing a variant of the phytase enzyme, said method comprising the following sequential steps: a) selecting at least one mother phytase enzyme, wherein the at least one mother phytase enzyme is a polypeptide selected from the group consisting of: a polypeptide comprising the amino acid sequence presented in SEQ ID No. 3 or an amino acid sequence that has an identity of at least 80% with it; a polypeptide that can be obtained from strain P1-29 of Buttiauxella sp. deposited with access number NCIMB 41248; a polypeptide that can be obtained by the expression of SEQ ID No. 2 or a nucleotide sequence that can be obtained from strain P1-29 of Buttiauxella sp. deposited with the accession number NCIMB 41248 or a nucleotide sequence having a sequence identity of at least 80% with it; and a polypeptide that can be obtained from strain P1-29 of Buttiauxella sp. deposited with the accession number NCIMB 41248 and wherein the peptide is obtained by the expression of SEQ ID No. 2 or a nucleotide sequence that can be obtained from strain P1-29 of Buttiauxella sp. deposited with the accession number NCIMB 41248 or of a nucleotide sequence having a sequence identity of at least 80% with it, wherein said polypeptide has phytase activity, wherein said polypeptide has a specific activity of at least 300 U / mg, wherein said specific activity is determined by incubating said polypeptide in a solution containing 2 mM phytate, 0.8 mM CaCl 2 in 200 mM sodium acetate buffer at pH 3.5, at a temperature from 37 ° C; b) generating at least one phytase variant by introducing at least one alteration of said mother phytase enzyme which is an insertion, a deletion or a substitution or combination thereof, of an amino acid residue in said mother phytase enzyme to obtain the less a variant of the phytase enzyme; c) scrutinize for said at least one variant of the phytase enzyme to identify an improved phytase enzyme variant which, in comparison to the mother phytase enzyme, has at least one or several improved properties selected from: i. greater thermostability; and / or ii. greater specific activity; and / or 30 iii. greater proteolytic stability; d) preparing said improved phytase enzyme variant |
