abstract | A gene expression modulation system comprising a) a first gene expression cassette that is capable of being expressed in a host cell comprising a polynucleotide encoding a first polypeptide comprising: i) a transactivation domain; ii) a domain of DNA binding that recognizes a response element associated with a gene whose expression is to be modulated; and ii) a Group B nuclear receptor ligand binding domain, in which Group B nuclear receptor ligand binding domain is selected from the group consisting of an Xretinoid receptor, a region II H binding protein. -2 (H-2RIIBP), a co-regulator-1 of Nuclear Receptors (RCoR-1), an ultra-spiracle protein, a 2C1 nuclear receptor and a ligand-binding domain of chorionic factor 1 (CF-1), and in the that amino acid residues that are in an equivalent or analogous position to that of amino acid residues 481, 482 and 483 of SEQ ID NO: 2 cause the gene expression modulation system to present a folding induction and an induction magnitude improved, compared to a gene expression modulation system that contains the corresponding wild-type Group B nuclear receptor ligand binding domain and b) a second gene expression cassette that is capable of expressing in a host cell comprising a polynucleotide encoding a second polypeptide comprising an alligator binding domain of nuclear receptors selected from the group consisting of an ecdysone receptor, a ubiquitous receptor, an orphan receptor 1, a nuclear steroid hormone receptor 1, a protein 15 that interacts with the retinoid X receptor, a liver X β receptor, a protein similar to the hormone steroid receptor, a liver X receptor, a liver X α receptor, a farnesoid X receptor, a protein 14 that interacts with receptors and a binding domain to farnesol receptor ligands. |
