Method for identifying a compound that activates a sirtuin, comprising: contacting a set of peptide substrates with a sirtuin in the presence of a test compound, in which the concentration of the peptide substrate in the set of peptide substrates is below the Km of the sirtuin for the peptide substrate, and in which the members of said set of peptide substrates comprise at least one acetylated lysine residue, and determine the level of acetylation of the set of peptide substrates by mass spectrometry, in which a reduction in the level of acetylation of the set of peptide substrates in the presence of the problem compound, compared to a control reaction in which the test compound is not included, is indicative of a compound that activates a sirtuin |
