abstract | The method for the purification of a monopegilated erythropoietin comprising the steps of providing a solution comprising monopegylated, polypegylated and non-pegylated erythropoietin, in which two steps of consecutive cation exchange chromatography are performed, and the recovery of the monopegylated erythropoietin in the second cation exchange chromatography step, characterized in that the same type of cation exchange material is used in both cation exchange chromatography steps and because the two consecutive cation exchange chromatography steps are performed using different elution methods, and in which the two consecutive steps of cation exchange chromatography comprise the following steps: a) applying a buffered aqueous solution comprising a mixture of monopegilated erythropoietin, non-pegylated polypegylated, and low molecular weight forms to a first chromatog column cation exchange raffia under conditions suitable for the binding of said monopegilated erythropoietin to the exchange material decationes contained in said first column, b) recovering the monopegilated erythropoietin from the first cation exchange chromatography column by a step elution method with an increase of the ionic strength of the elution buffer, in which the fraction of said monopegylated erythropoietin in the recovered solution increases compared to the mixture applied, c) apply the monopegilated erythropoietin recovered from step b) in a second column of low cation exchange chromatography the suitable conditions for the attachment of said cation exchange monopegilated erythropoietin contained in said second column, wherein the decation exchange material contained in said second column is the same type as the cation exchange material in the first column, d ) recover r purified monopegilated erythropoietin in a substantially homogeneous form from the second column of cation exchange chromatography by a continuous elution method with a continuous increase in the ionic strength of the elution buffer. |
